Sykehistorien gir ofte mistanke om diagnosen, men den er utilstrekkelig til å stille en sikker diagnose. I to studier fant man at bare 1 av 10 som oppga å ha lateksallergi, i realiteten hadde det.

De to mest aktuelle testene på lateksallergi er blodprøve med måling av spesifikke lateksallergener (IgE) og hudprikktest. Hudprikktesten er den mest følsomme og foretrukne metoden.
